Pantropic canine coronavirus induces canine M1 macrophage polarization in vitro

چکیده
Emerging coronavirus infections are a major threat to global public health. In this respect, novel recombination of canine (CCoV) and feline (FCoV) was described among human biological samples, giving rise potential zoonosis. Despite all efforts, the host‒virus immune response related CCoV is missing. study, pantropic infection macrophages derived from peripheral blood monocytes performed. After infection, were first polarized M1 and/or M2 phenotype. Moreover, kinetics, cell viability, apoptosis, mitochondrial dysfunction associated with reactive oxygen species oxide nitric production measured. Our results demonstrated that virus mainly host classically activated (M1) phenotype, as by amoeboid morphology numerous fibrillary cytoplasmic processes followed classical phenotypes. Viral released new particles at 18 h post-infection decrease in viable cells. Furthermore, upon cells exhibited reduced phagocytosis properties, evidenced neutral red uptake assay. This vitro method open an avenue for further studies on host-virus interaction.

European surveillance for pantropic canine coronavirus.
Highly virulent pantropic canine coronavirus (CCoV) strains belonging to subtype IIa were recently identified in dogs. To assess the distribution of such strains in Europe, tissue samples were collected from 354 dogs that had died after displaying systemic disease in France (n = 92), Hungary (n = 75), Italy (n = 69), Greece (n = 87), The Netherlands (n = 27), Belgium (n = 4), and Bulgaria (n = ...
متن کاملFull-Genome Sequence of Pantropic Canine Coronavirus
Pantropic canine coronavirus (CCoV) was first detected in young dogs in Italy in 2005, but the complete genome sequence of this virus had not yet been determined. Here, we report the full-length genome sequence of the prototype strain CB/05, which showed that this virus is genetically similar to CCoV-IIa viruses.
متن کاملHepcidin Induces M1 Macrophage Polarization in Monocytes or THP-1 Derived Macrophages
Background: Macrophage polarization plays a critical role in determining the inflammatory states. Hepcidin is a key negative regulator of iron homeostasis and functions. Although hepcidin has been shown to affect ferroportin expression in macrophages, whether it affects macrophage polarization is still largely unknown. Objective: To address whether hepcidin ind...
متن کاملAntigenic detection of Canine Coronavirus in diarrheic dogs in Ahvaz
Abstract: The Purpose of this study was to evaluate the prevalence of Canine Coronavirus (CCV) in diarrheic dogs of Ahvaz district. Fecal samples were collected from 58 diarrheic dogs between 2005 and 2006. Dogs were grouped by age (less than and above 6 months), sex, breed, geographical location (urban and rural) and quality of diarrhea (hemorrhagic and non-hemorrhagic) to determine whether th...
متن کاملPrevalence of Canine Monocytic Ehrlichiosis in Canine Population across India
Canine ehrlichiosis is a very important emerging disease in India. This study is the first attempt screening a large number of canines in India for the detection of canine monocytic ehrlichiosis. In the present study, 510 blood samples of dogs were screened for the presence of Ehrlichia canis and other variants of Anaplasmataceae family by serological and molecular methods.Out of the 510 serum ...
